CJC-1295 + Ipamorelin Blend

Also known as CJC-1295 + Ipamorelin · CJC/Ipa · GHRH + GHRP blend

The canonical GH-axis blend — two receptors, one synergy argument

Last reviewed · Educational reference — not medical advice.

Half-life
Not characterised in published human pharmacokinetic studies
Route
Subcutaneous injection in reported research use
Status
Compounded blend — no controlled studies of the combination located
Mechanism
Dual-receptor stimulation of GH release: CJC-1295 acts as a GHRH analogue on the stimulatory arm, while ipamorelin acts at the ghrelin/GHS receptor and reduces somatostatin inhibitory tone. The combination is proposed to be more than additive because it simultaneously increases stimulation and reduces inhibition.

Overview

CJC-1295 with ipamorelin is the most widely used growth hormone axis blend, and unlike most blends its rationale rests on an actual physiological argument rather than simple co-packaging.

Growth hormone release from the pituitary is governed by two opposing systems: GHRH stimulates it, and somatostatin inhibits it. CJC-1295 is a GHRH analogue acting on the stimulatory arm. Ipamorelin is a selective ghrelin receptor (GHS-R) agonist acting through a separate receptor, and ghrelin receptor agonists also suppress somatostatin tone. Combining them therefore pushes on the accelerator and releases the brake simultaneously, which is the basis for expecting more than additive GH release.

Ipamorelin's selectivity is the reason it is preferred over older GHRPs in these blends: GHRP-6 and GHRP-2 also stimulate cortisol and prolactin release, whereas ipamorelin was designed for a cleaner profile.

A critical distinction is often lost in vendor listings:

CJC-1295 with DAC and CJC-1295 without DAC are pharmacologically different . The DAC (drug affinity complex) version binds albumin and produces sustained elevation lasting days; the no-DAC version, also called Modified GRF 1-29, is short-acting and preserves the pulsatile GH release pattern. Since GH is normally secreted in pulses and continuous elevation is not the physiological state, this is a substantive difference, not a convenience one. Blends should specify which version they contain.

Limitations & open questions

This is a fixed-ratio premixed combination, and that carries specific consequences beyond those of its individual components. The components cannot be titrated independently — adjusting one means adjusting all of them. If a reaction occurs, attribution is impossible. And the combination itself has not been studied: evidence for the individual compounds does not transfer to the mixture, because combined pharmacology is not the sum of separate pharmacologies. Blends are a convenience and pricing format, not a validated pharmacological unit. A blend-specific hazard applies here: 'CJC-1295' is ambiguous between the DAC and no-DAC forms, which differ in duration from days to minutes and therefore in whether GH release remains pulsatile. A blend that does not specify which form it contains cannot be meaningfully evaluated. Raising GH and IGF-1 also has its own risk profile, including insulin resistance and effects on tissue growth.

Pharmacology

A mixture rather than a single chemical entity. The pharmacokinetics depend critically on which CJC-1295 form is used: the DAC version binds albumin for sustained multi-day elevation, while the no-DAC version (Modified GRF 1-29) is cleared within minutes. Ipamorelin is short-acting. See the individual entries.

Frequently asked questions

Why combine a GHRH analogue with a ghrelin receptor agonist?

They act on different receptors and different sides of the control system. GHRH stimulates GH release; ghrelin receptor agonists stimulate it *and* reduce somatostatin's inhibitory brake. Pushing the accelerator while easing the brake is the synergy argument.

Does it matter whether the blend uses CJC-1295 with or without DAC?

Substantially. With DAC it binds albumin and elevates GH for days; without DAC (Modified GRF 1-29) it is cleared in minutes and preserves pulsatile release. GH is normally pulsatile, so this is a real pharmacological difference, and any blend should state which form it contains.

Why ipamorelin rather than GHRP-6 or GHRP-2?

Selectivity. GHRP-6 and GHRP-2 also stimulate cortisol and prolactin release; ipamorelin was designed for a cleaner profile at the GHS receptor.
