Why Titrate Slowly?
The standard ladder

A common schedule is 0.25 → 0.5 → 1.0 → 1.7 → 2.4 mg weekly, holding each step about four weeks. The starting 0.25 mg is generally a tolerance-building dose.

Fewer side effects

Gradual escalation reduces gastrointestinal side effects — nausea is the most common — by giving the body time to adjust before each increase.

Flexible hold times

Around four weeks per step is typical, but the hold can be extended if a step isn't well tolerated.

Converting mg to Syringe Units
Why conversion matters

Semaglutide vials are reconstituted with bacteriostatic water, and your dose is drawn on a U-100 insulin syringe marked in units.

The formula

Concentration (mg/mL) = vial mg ÷ water mL. Units = (dose mg ÷ concentration) × 100.

Use the dose converter

Enter your vial size and water volume in the reconstitution calculator to convert every mg step into exact units for your vial.

Tip: Start at the lowest dose and titrate slowly. Do not exceed the recommended titration schedule.

Important Notes

Semaglutide requires a prescription. This calculator illustrates a standard FDA-based titration schedule for educational and research purposes only — it is not medical advice or a prescribing tool. Consult a healthcare professional for actual dosing decisions.
